چکیده

Introduction Patients with unexplained recurrent febrile episodes and CASP1 variants suffer from systemic sterile inflammation despite altered enzymatic activity of procaspase-1 and reduced IL-1b release. Most recent findings from our group indicate that the proinflammatory effects of CASP1 variants with reduced or abrogated enzymatic activity could be due to receptor interacting protein kinase 2 (RIP2) mediated increase of NF-kB activation. These findings are additionally supported by a trend to elevated IL-6 and TNF-a expression in patients with CASP1 variants.

Curcumin: a double hit on malignant mesothelioma.

Inflammation is a key mediator in the development of malignant mesothelioma, which has a dismal prognosis and poor therapeutic strategies. Curcumin, a naturally occurring polyphenol in turmeric, has been shown to possess anticarcinogenic properties through its anti-inflammatory effects. Inflammasomes, a component of inflammation, control the activation of caspase-1 leading to pyroptosis and pro...

Characterisation of human iPS cells harbouring the p.A329T variant of caspase-1

Introduction Naturally occurring genetic variants of the CASP1 gene are associated with autoinflammation in patients suffering from recurrent febrile episodes and generalized inflammation. The resulting caspase-1 variants have reduced enzymatic activity due to destabilization of the caspase-1 tetramer. In addition, autoprocessing of the pro-caspase-1 variants is reduced and hence, CARDCARD inte...

Enzymatically inactive procaspase-1 stabilizes the ASC-pyroptosome

Caspase-1 is a key player during the initiation of pro-inflammatory innate immune responses, activating pro-IL-1β in so-called inflammasomes. A subset of patients with recurrent febrile episodes and systemic inflammation of unknown origin harbor mutations in CASP1 encoding caspase-1. CASP1 variants result in reduced enzymatic activity of caspase-1 and impaired IL-1β secretion. The apparent para...

Radiological dose assessment of naturally occurring radioactive materials generated by the petroleum industry in wildlife: A case study of chinkaras of Lavan Island, Iran

Human activities such as oil and gas production can enhance the natural level of naturally occurring radioactive materials (NORM) in by-product and waste streams. Iran has been among the top five oil producing countries since 2005. This high production rate emphasizes the importance of NORM management to ensure the safety of humans and wildlife. Petroleum storage and transport facilities are lo...

Preparation and In Vitro Evaluation of Sustained-Release Matrix Tablets of Flutamide Using Synthetic and Naturally Occurring Polymers

Frequent dosing of the potent anti-androgen, flutamide, is necessary to reach a therapeutic level for the treatment of prostatic carcinoma. Sustained delivery of the drug could reduce the adverse effects such as gastrointestinal disorders and improve patient compliance. In the present study sustained-release matrix tablets of flutamide were prepared by direct compression method using different ...
